A column of prepared Dowex can be used to remove ethidium from DNA solutions, such as DNA bands collected off CsCl gradients. |

1. Obtain 1 kg (two 500 g bottles) of Dowex Ag50W-X8 200-400 mesh.
2. Soak the resin in 4 liters of 1 M HCl for 15 min while mixing on a stir plate.
3. Pour the resin slurry into a scintered glass funnel and run 500 ml of ddH2O through the resin five times.
4. Place the resin into a 5-liter vessel and soak the resin in 4 liters of 1 M NaOH for 15 min with stirring.
5. Add the resin back to the scintered glass funnel and run 500 ml of ddH2O through the resin five times.
6. Add 500 ml of ddH2O and adjust the slurry pH to 6.0 with HCl.
7. Autoclave the resin and store it at 4°C.
8. Prior to use, equilibrate the resin in a column with TEN.
